5-Amino-1MQ is a selective small-molecule inhibitor of NNMT, an enzyme implicated in adipose-tissue metabolic dysregulation. NNMT consumes the methyl group donor SAM and the NAD⁺ precursor nicotinamide, creating a metabolic sink that depletes cellular methylation capacity and NAD⁺ pools in tissues with high NNMT expression. Selective inhibition aims to restore both pools.
Selective NNMT inhibition (IC₅₀ ≈ 1.5 µM) Restores NAD⁺ salvage in NNMT-high tissues Preclinical adipose-tissue & muscle regeneration data Oral bioavailability established in animal models
5-Amino-1MQ competitively inhibits NNMT (IC₅₀ ≈ 1.5 µM), reducing 1-methylnicotinamide accumulation and restoring nicotinamide availability for NAD⁺ salvage. In white-adipose-tissue models the result is enhanced lipolysis, improved insulin sensitivity, and modulation of the methyl/NAD⁺ balance toward a phenotype associated with leaner metabolic profile.
Preclinical models of NNMT-mediated adipose dysregulation. Investigates whether restoring NAD⁺ salvage improves metabolic phenotype.
Limited human safety data. Iodide salt form contributes iodine load — protocols in iodine-replete subjects should monitor thyroid function over extended cycles. Long-term selective inhibition has not been characterized in human research; standard research-compound caution applies.
