5-Amino-1MQ is a selective small-molecule inhibitor of NNMT, an enzyme implicated in adipose-tissue metabolic dysregulation. NNMT consumes the methyl group donor SAM and the NAD⁺ precursor nicotinamide, creating a metabolic sink that depletes cellular methylation capacity and NAD⁺ pools in tissues with high NNMT expression. Selective inhibition aims to restore both pools.

In white-adipose-tissue models the result is enhanced lipolysis, improved insulin sensitivity, and modulation of the methyl/NAD⁺ balance toward a phenotype associated with leaner metabolic profile.
