Somatropin (191AA) is a recombinant form of human growth hormone with the full 191-amino acid sequence, identical to endogenous pituitary . Production is via rDNA technology in Escherichia coli — Genentech's pioneering work (1981) made somatropin the first commercial rDNA pharmaceutical. Mechanistically binds GHR ( receptor) on the cell surface, dimerizes it and activates the JAK2/STAT5 signaling cascade. STAT5 induces synthesis in the liver — is the secondary mediator responsible for anabolic and anti-apoptotic effects in peripheral tissues. Beyond the pathway, has direct effects: lipolysis in adipocytes via hormone-sensitive lipase (especially visceral), hepatic glycogenolysis and stimulation of chondrocytes in epiphyseal plates. Rudman's classic study (NEJM 1990) documents body composition redistribution in older men after 6 months of therapy. Unlike / peptides that stimulate endogenous release, somatropin bypasses the pituitary and delivers exogenous directly. This is a stronger but less physiological signal. Half-life s.c. ~3.5 hours. Storage: 2-8°C lyophilized; reconstituted — 14 days at 2-8°C, protected from light and without freezing.
