What is Tirzepatide?
Tirzepatide combines GIP and receptor agonism in a single 39-amino-acid molecule with a 20-carbon fatty acid moiety for albumin binding (~5-day half-life). The dual mechanism activates both incretin axes simultaneously — a strategy validated by SURPASS (T2D) and SURMOUNT (obesity) Phase 3 programs. SURMOUNT-1 (NEJM 2022) reported 22.5% mean weight loss at 15 mg/week, the largest effect size yet documented for pharmacological weight reduction.
